If you dont care about a customized message, I think the simplest way would be to change the mailbox properties and enable the "Require all senders are authenticated" option. Other ways you could do the same is powershell script a response to modify the mailboxes Out of office message, or create a inbox rule that would auto - respond with a customized message. If you want to customize a message response you can do what jrp87 said, and use a transport rule to respond to external email sent to a person or group of people. It will "disable" by actually deleting the mailbox after whatever time period your environment is set to, 30 days if left at default. This only started being a problem in the last two day.
You can't disable the way you would disable an AD account. I'm having a weird issue with my Outlook, 365, Exchange account in Mac Mail.
